Coombs Test – Direct (DCT), EDTA Blood
Direct Coombs Test (DCT)
The Direct Coombs Test (DCT), also known as the Direct Antiglobulin Test (DAT), is a blood test used to detect antibodies or complement proteins attached to the surface of red blood cells (RBCs). This test helps diagnose conditions where the immune system mistakenly destroys red blood cells, leading to hemolytic anemia.
Why is this Test Done?
The DCT test is commonly recommended to:
- Diagnose autoimmune hemolytic anemia
- Investigate unexplained anemia or jaundice
- Detect transfusion reactions
- Evaluate hemolytic disease in newborns
- Monitor certain autoimmune or infectious conditions
Sample Type
Preparation
- No fasting or special preparation is usually required.
Interpretation
-
Positive DCT: Indicates antibodies or complement proteins are attached to red blood cells.
-
Negative DCT: No significant antibody coating detected on RBCs.
Symptoms That May Require This Test
- Fatigue and weakness
- Pale skin
- Jaundice
- Dark urine
- Shortness of breath
- Rapid heartbeat
Test Benefits
- Helps identify immune-related destruction of red blood cells
- Supports diagnosis of hemolytic disorders
- Assists in blood transfusion evaluation
- Useful in newborn screening for hemolytic disease
